Breast cancer is the most common diagnosed cancer among women and the second only after lung cancer as the primary cause of cancer deaths in women. Conjugated linoleic acid (CLA), a naturally occurring compound, found in ruminants products has been shown to possess anti-cancer ability in in vivo and in vitro. Our research interest is to examine the mechanisms underlying the anti-mammary gland tumor effects of CLA. In our study, we have first shown that CLA is able to up-regulate the estrogen-regulated tumor suppressor gene, protein tyrosine phosphatase gamma (PTPgamma). PTPgamma has been proved to regulate cell cycle progression and delay cell cycle reentry. Increasing expression of PTPγ by CLA treatment indicates the potential anti-cancerous role of CLA for human breast cancer. Our study also shown that CLA modulates PGE2 signaling pathway, alters breast microenvironment, and induces apoptosis in mammary gland tumor in in vitro models. Among the endocrine factors associated with breast cancer, estrogens are considered to play a central role in human breast carcinogenesis. The anti-estrogen tamoxifen is the most frequently prescribed drug for estrogen responsive breast cancer patients and also is recommended for women at high risk of developing breast cancer. Our study first demonstrates that CLA potentiates the anti-estrogenic effects of Tamoxifen (Tam) in human breast cancer cells. Collectively, these studies add to a growing evidence of applying CLA as a naturally occurring anti-cancerous agent. The investigations not only extend our knowledge of the mechanisms of how CLA exerts anti-mammary gland tumor activities but also provide rationales of using CLA daily consumption for the preventive and therapeutic purpose for human and dog mammary gland tumors.
